This will unlock when you “rescue at least 10 prisoners in one Metal Slug 6 mission.”
The easiest mission to do this is in the first one. Play cautiously so that you don’t get killed. If you do get killed, then you will lose all of your rescued prisoners.
You will be able to rescue up to 15 prisoners on the first mission although you only need 10 to unlock this trophy. You should find them easily on the main path as long as you don’t die.
When you encounter the boss, stay on the left edge of the screen to avoid most of his attacks. You should then only worry about the rock throws.

Make sure you set the Continue setting to Free Play, which should grant you unlimited lives. You can also lower the difficulty if you want. There is no save option, so you need to beat the game in one go.

For “Mission Complete!”, you need to “finish the game on Difficulty HARD (Must play through all stages).”
For “Speedster: Hard” you need to “reach Rank 1st RECORD TIME on HARD difficulty. Must play through all stages without quitting game.”
To unlock both, you need to beat the game by playing in Hard Difficulty.
You should be able to get the Speedster: Hard trophy/achievement no matter what since the Clear Time tops out at 59 minutes and 59 seconds, so you will be tied for first even if your run lasts longer.
